Lentiviral Vector Purification: Optimizing the Downstream Chromatography Step

by Ascend Advanced Therapies

In this presentation from ASGCT 2026, Shanshan Zhu shares new insights into optimizing anion exchange chromatography for lentiviral vector (LVV) purification.

Strong anion exchange chromatography is widely used in downstream processing, but recovery can vary depending on the composition of the feed stream. Shanshan’s work demonstrates that host cell protein levels play a critical role: if the feed is too clean, vectors may bind too tightly to the resin; if impurity levels are too high, host cell proteins can compete for binding sites.

By understanding this balance, Ascend is developing more robust and scalable purification processes that improve consistency and recovery in lentiviral vector manufacturing.

This presentation highlights the importance of process understanding in building reliable downstream strategies for gene therapy production.
